This anti-PD-1 antibody is a monoclonal antibody being evaluated by the Chinese PLA General Hospital (PLAGH) in a Phase I/II clinical trial for the treatment of locally advanced or metastatic pancreatic cancer. It is a key component of a pan-immunotherapy regimen that combines PD-1 blockade with manganese chloride (MnCl2) and standard chemotherapy (nab-paclitaxel and gemcitabine). The antibody functions as a checkpoint inhibitor by binding to the programmed cell death protein 1 (PD-1) receptor on T-cells, thereby preventing its interaction with the ligands PD-L1 and PD-L2 on tumor cells. This action restores the anti-tumor activity of T-cells that has been suppressed by the tumor's immune evasion mechanisms. The trial specifically investigates the use of manganese as an immunoadjuvant to activate the cGAS-STING pathway, which is intended to enhance the efficacy of the PD-1 inhibitor in the typically cold immune environment of pancreatic adenocarcinoma.
