This novel antibody-drug conjugate (ADC) is an experimental therapeutic platform designed to overcome resistance to immune checkpoint inhibitors by targeting the extracellular matrix (ECM) remodeling process. It consists of an anti-PD-1 monoclonal antibody backbone conjugated to a selective enzymatic inhibitor of Bone Morphogenetic Protein 1 (BMP1). While anti-PD-1 therapy typically activates CD8+ T cells, research has shown that these cells also secrete BMP1, a metalloprotease that drives pathological collagen deposition and immune exclusion. By delivering the BMP1 inhibitor directly to activated T cells via the PD-1 targeting arm, the ADC prevents therapy-induced ECM stiffening and enhances the infiltration and efficacy of cytotoxic T cells. Preclinical studies in murine models of melanoma, breast, and lung cancer have demonstrated that this dual-mechanism construct improves tumor control compared to conventional anti-PD-1 therapy.
