Anti-PD-1 scFv (single-chain variable fragment) refers to a class of recombinant antibody fragments designed to target the Programmed Cell Death Protein 1 (PD-1) receptor. These molecules consist of the variable heavy (VH) and light (VL) chains of an anti-PD-1 antibody joined by a flexible peptide linker. By binding to PD-1, the scFv blocks the interaction with its ligands, PD-L1 and PD-L2, thereby alleviating the immunosuppression typically found in the tumor microenvironment and reinvigorating exhausted T cells. This modality is particularly valued for its small molecular weight (~27 kDa), which facilitates better tumor tissue penetration compared to full-length monoclonal antibodies like nivolumab or pembrolizumab. It is frequently employed in "armored" CAR-T cell therapies, where the T cells are engineered to secrete the scFv locally, or as a payload in circular RNA and nanoparticle delivery systems to minimize systemic toxicity and immune-related adverse events.
