anti-PD-L1-γ1 is an immune checkpoint inhibitor, specifically an anti-PD-L1 antibody, delivered through an in vivo hematopoietic stem/progenitor cell (HSPC) gene therapy approach. This innovative method involves genetically modifying HSPCs to express the anti-PD-L1-γ1 effector gene only when recruited to and differentiated by a tumor. This early, self-activating expression aims to overcome the immunosuppressive tumor microenvironment, providing immunoprophylaxis against cancers. It has shown efficacy in preclinical models of mouse mammary carcinoma and ovarian cancer, indicating its potential for clinical translation in high-risk germline mutation carriers.
