Overview

anti-PD-L1 mAb-ODN conjugate is a preclinical-stage immunotherapeutic conjugate consisting of an anti-programmed death-ligand 1 (PD-L1) monoclonal antibody covalently linked to a synthetic oligodeoxynucleotide (ODN) containing unmethylated CpG motifs, which acts as a Toll-like receptor 9 (TLR9) agonist. Developed by researchers at the University of Miami, this conjugate is designed to simultaneously block the PD-1/PD-L1 inhibitory pathway and activate innate and adaptive immune responses via TLR9 stimulation in the tumor microenvironment. In preclinical murine models of breast cancer (4T1), the conjugate demonstrated significant tumor growth inhibition and enhanced splenocyte proliferation compared to non-conjugated controls.
