Overview

**Anti-PD-L2** refers to monoclonal antibodies or other inhibitory agents that specifically target **Programmed Death Ligand 2 (PD-L2)**, an immune checkpoint molecule and ligand of PD-1. PD-L2 regulates T-cell activation by engaging with PD-1 and suppressing immune responses in peripheral tissues and within the tumor microenvironment. Blocking PD-L2 can unleash anti-tumor T-cell activity, and anti-PD-L2 agents are being investigated as monotherapy or in combination with anti-PD-L1/PD-1 therapies to overcome immune resistance, especially in cancers where PD-L2 is highly expressed. PD-L2 has a higher binding affinity for PD-1 than PD-L1 and can be upregulated in tumor cells, dendritic cells, and macrophages, modulating immune escape. Anti-PD-L2 therapies may hold potential to complement or enhance the effectiveness of current PD-1/PD-L1 blockade approaches in cancer immunotherapy[2][4][5][7].
