**anti-PD1-MDNA132** is a preclinical Bi-functional SuperKine ImmunoTherapy developed by Medicenna. It is a single engineered fusion protein comprising an anti-programmed cell death protein 1 antibody linked to MDNA132, an engineered interleukin-13 superkine that selectively binds the tumor-associated interleukin-13 receptor subunit alpha-2 without binding interleukin-13 receptor subunit alpha-1. The construct is designed to anchor PD-1 checkpoint blockade within interleukin-13 receptor subunit alpha-2-positive tumor microenvironments while retaining anti-PD-1-mediated PD-1/programmed death-ligand 1 blockade. In the reported 2023 AACR studies, it retained selective binding to PD-1 and interleukin-13 receptor subunit alpha-2 and showed PD-1/programmed death-ligand 1 blockade comparable to the parental anti-PD-1 antibody.
