Overview

anti-PD1H refers to a class of monoclonal antibodies designed to target and inhibit the Programmed Death-1 Homolog (PD1H), more commonly known as V-domain Ig suppressor of T cell activation (VISTA). PD1H is an immune checkpoint protein and a member of the B7 family, primarily expressed on myeloid-derived suppressor cells (MDSCs), regulatory T cells (Tregs), and other tumor-infiltrating lymphocytes. It functions as a negative regulator of T-cell activation and cytokine production. By blocking PD1H, these antibodies aim to overcome immune suppression within the tumor microenvironment, particularly in 'cold' tumors like pancreatic ductal adenocarcinoma (PDAC). Research, such as that conducted in KPC mouse models, suggests that anti-PD1H therapy may be more effective when combined with epigenetic modulators like decitabine (DAC), which can polarize macrophages toward an M1 phenotype and increase CD8+ T-cell infiltration.
