Overview

Anti-PDPN antibodies are monoclonal antibodies designed to target podoplanin (PDPN), a type I transmembrane sialoglycoprotein frequently overexpressed in various malignancies, including glioblastoma, mesothelioma, and squamous cell carcinomas. PDPN plays a critical role in the tumor microenvironment (TME) by interacting with receptors such as CLEC5A on tumor-associated macrophages (TAMs). This interaction triggers Syk-JAK-STAT3 signaling, which drives the polarization of TAMs toward an immunosuppressive phenotype, thereby promoting tumor growth and immune evasion. By binding to PDPN, these antibodies block its interaction with CLEC5A, inhibiting TAM-mediated immunosuppression and delaying tumor progression. Research presented at SNO 2024 demonstrates that pharmacological targeting of PDPN with blocking antibodies can suppress glioma growth and prolong survival in preclinical models.
