Drug intelligence / Profile preview
Anti-PfRH5 monoclonal antibodies are a group of research-stage monoclonal antibodies targeting the *Plasmodium falciparum* reticulocyte-binding protein homolog 5 (PfRH5), an essential blood-stage parasite adhesin. PfRH5 forms a trimeric complex with cysteine-rich protective antigen (PfCyRPA) and PfRH5-interacting protein (PfRIPR) to bind the host erythrocyte receptor basigin (CD147), a critical step for merozoite invasion. By binding to PfRH5, these antibodies block its interaction with basigin, thereby neutralizing merozoite invasion and preventing or treating blood-stage malaria. Various academic institutions have developed different antibody clones (such as 5A08, c2AC7, and c4BA7) to evaluate their neutralizing capacity, map protective epitopes, and guide next-generation malaria vaccine design.
