Anti-podoplanin antibodies are monoclonal antibodies that target podoplanin (PDPN), a type I transmembrane sialoglycoprotein also known as Aggrus. Podoplanin is highly expressed in various malignancies, including malignant mesothelioma, glioblastoma, and squamous cell carcinomas, where it plays a critical role in tumor-induced platelet aggregation and metastasis by interacting with the C-type lectin-like receptor 2 (CLEC-2) on platelets. In clinical research, these antibodies are utilized in diagnostic microfluidic platforms, such as the Universal CTC-chip, to capture circulating tumor cells (CTCs) from the blood of patients with lung cancer or mesothelioma. Therapeutically, anti-podoplanin antibodies are being investigated as naked antibodies, antibody-drug conjugates (ADCs), and chimeric antigen receptor (CAR) T-cell therapies to inhibit tumor progression and hematogenous metastasis.
