Overview

The anti-PODXL monoclonal antibody is a therapeutic agent targeting Podocalyxin (PODXL), a CD34-related sialomucin and embryonic stem cell marker that is overexpressed in various aggressive malignancies, including glioblastoma, breast cancer, and colorectal cancer. While PODXL is also expressed in normal tissues such as vascular endothelial cells and renal podocytes, this specific antibody was developed using CasMab technology to achieve cancer-specific binding, thereby minimizing potential off-target toxicity. The antibody, particularly the clone PcMab-47 and its mouse-human chimeric derivatives, exerts anti-tumor activity primarily through antibody-dependent cellular cytotoxicity (ADCC). Preclinical studies have demonstrated its efficacy in inhibiting tumor growth in xenograft models of PODXL-expressing cancers.
