Overview

Anti-PODXL neutralizing antibodies are monoclonal antibodies designed to target and inhibit podocalyxin (PODXL), a sialylated transmembrane glycoprotein belonging to the CD34 family. PODXL is highly expressed in various cancers, including triple-negative breast cancer (TNBC), pancreatic ductal adenocarcinoma, and colorectal cancer, where it plays a key role in cell adhesion, cancer stemness, and the formation of circulating tumor cell (CTC) clusters. In preclinical models of TNBC, chemotherapy evasion (such as with paclitaxel) leads to the loss of ST6GAL1-catalyzed sialylation, which promotes PODXL-mediated CTC clustering and subsequent lung metastasis. Neutralizing antibodies against PODXL block homotypic tumor cell aggregation and inhibit metastatic seeding, presenting a potential therapeutic strategy to overcome chemotherapy resistance and prevent metastasis.
