Anti-programmed death 1 (PD-1) antibodies are a class of monoclonal antibodies that function as immune checkpoint inhibitors. They are designed to bind to the PD-1 receptor, a protein found on the surface of T cells and other immune cells. Under normal conditions, PD-1 acts as a brake on the immune system to prevent overactivation and maintain self-tolerance. However, many cancer cells exploit this pathway by expressing ligands (PD-L1 and PD-L2) that bind to PD-1, effectively silencing the T-cell response and allowing the tumor to evade immune detection. By blocking this interaction, anti-PD-1 antibodies restore the immune system's ability to recognize and destroy malignant cells. This class of therapy has become a cornerstone in the treatment of numerous cancers, including melanoma, non-small cell lung cancer, and urothelial carcinoma, and is used both as monotherapy and in combination with other antineoplastic agents.
