Overview

Anti-programmed death-ligand 1 saporin (anti-PD-L1-SAP) is an experimental immunotoxin designed to target and eliminate cells expressing PD-L1 (CD274). The drug consists of a monoclonal antibody specific for PD-L1 conjugated to saporin, a type II ribosome-inactivating protein (RIP) derived from the plant *Saponaria officinalis*. Upon binding to PD-L1 on the surface of cancer cells or immunosuppressive cells within the tumor microenvironment, the conjugate is internalized via endocytosis. Saporin then facilitates its own escape into the cytosol—a process that can be significantly enhanced by **photochemical internalization (PCI)**, a light-controlled technology that triggers endosomal release. Once in the cytoplasm, saporin acts as an RNA N-glycosylase, irreversibly inactivating the 28S ribosomal RNA of the 60S subunit, which halts protein synthesis and leads to apoptotic cell death. This therapeutic approach is being explored for the treatment of PD-L1-positive malignancies, including triple-negative breast cancer and non-small cell lung cancer.
