Drug intelligence / Profile preview
anti-PROS1 refers to monoclonal or polyclonal antibodies designed to target and neutralize Protein S (PROS1), a vitamin K-dependent plasma glycoprotein. PROS1 plays a dual role in human physiology: it acts as an anticoagulant cofactor for activated protein C (aPC) and tissue factor pathway inhibitor alpha (TFPIα), and it serves as a high-affinity ligand for the TAM (TYRO3, AXL, and MERTK) family of receptor tyrosine kinases. In hematology, therapeutic anti-PROS1 antibodies (such as latarcibart/VGA-039) are developed to promote thrombin generation and restore hemostasis, offering a potential subcutaneous prophylactic treatment for bleeding disorders like von Willebrand disease (VWD). In oncology, neutralizing PROS1 is investigated to disrupt the PROS1-MERTK or PROS1-AXL signaling pathways, which are implicated in tumor cell proliferation, migration, and the polarization of tumor-associated macrophages (TAMs) toward an immunosuppressive phenotype in the tumor microenvironment.
