anti-PSGL-1 antibody refers to a class of monoclonal antibodies, such as Verseau Therapeutics' lead candidate VTX-0811, that target P-selectin glycoprotein ligand-1 (PSGL-1). PSGL-1 is an immune checkpoint molecule highly expressed on immunosuppressive tumor-associated macrophages (TAMs) and M2-polarized macrophages within the tumor microenvironment. By binding to and blocking PSGL-1, these antibodies facilitate the repolarization of macrophages from a pro-tumorigenic M2-like state to a pro-inflammatory M1-like state. This phenotypic shift is driven by the upregulation of TNF-α/NF-κB and chemokine signaling pathways and the downregulation of metabolic pathways like oxidative phosphorylation and fatty acid metabolism. Preclinical studies have demonstrated that anti-PSGL-1 antibodies can reduce suppressive myeloid populations, increase the infiltration of effector CD8+ T cells, and suppress tumor growth in models such as melanoma, particularly when used in combination with PD-1 inhibitors.
