Drug intelligence / Profile preview
An anti-PSGL1 antibody is a monoclonal antibody designed to target P-selectin glycoprotein ligand-1 (PSGL-1, also known as CD162 or SELPLG). PSGL-1 is a mucin-like glycoprotein expressed on the surface of most hematopoietic cells, including leukocytes and macrophages, and acts as a high-affinity counter-receptor for P-, E-, and L-selectins. It plays a critical role in leukocyte trafficking, cell adhesion, and immune regulation. In oncology, PSGL-1 functions as an immune checkpoint on T cells and is highly expressed on immunosuppressive tumor-associated macrophages (TAMs). Blocking PSGL-1 can reprogram TAMs from an M2-suppressive phenotype to an M1-inflammatory phenotype, activate T cells, and enhance anti-tumor immune responses. Conversely, agonistic anti-PSGL-1 antibodies are being developed to downregulate pathogenic T cells for the treatment of autoimmune and inflammatory diseases.
