anti-pTα-DM1 is a preclinical antibody-drug conjugate (ADC) consisting of a monoclonal antibody (clone K5G3) targeting the invariant pTα subunit of the human pre-T cell receptor (pre-TCR), conjugated to the cytotoxic microtubule inhibitor mertansine (DM1) via a non-cleavable MCC linker. The pre-TCR is a developmental checkpoint receptor expressed transiently in developing T-cell progenitors but absent on mature healthy T cells, making it a highly specific biomarker for approximately 50% of T-cell acute lymphoblastic leukemia (T-ALL) cases. By targeting pTα, the ADC is internalized and delivers its cytotoxic payload to leukemia-initiating cells (LICs), effectively inhibiting tumor progression and prolonging survival in patient-derived xenograft (PDX) models of relapsed/refractory T-ALL. Preclinical studies have demonstrated that this targeted approach selectively impairs malignant cell proliferation while sparing normal mature T-cell populations.
