Overview

Anti-pTα monoclonal antibody is a biologic therapeutic agent that targets the invariant pTα (pre-T cell receptor alpha) subunit of the human pre-T cell receptor (pre-TCR). It is specifically designed to target leukemia-initiating cells (LICs) in T-cell acute lymphoblastic leukemia (T-ALL), particularly in the relapsed or refractory setting. The pre-TCR is a surface receptor essential for T-cell development and is expressed on LICs in human T-ALL but has limited expression on normal mature T cells, making it a promising biomarker for selective immunotherapy. Preclinical studies using the research clone K5G3, both as a standalone monoclonal antibody and as an antibody-drug conjugate (ADC), have demonstrated significant inhibition of leukemic cell engraftment and delayed tumor progression in mouse xenograft models.
