The anti-PTHrP vaccine is an investigational therapeutic peptide vaccine developed by Pepscan Therapeutics. It utilizes Pepscan's proprietary **CLIPS** (Chemical Linkage of Peptides onto Scaffolds) technology to create conformationally constrained peptides that mimic the structure of parathyroid hormone-related protein (PTHrP). The vaccine is designed to treat **humoral hypercalcemia of malignancy (HHM)**, a paraneoplastic syndrome where tumors secrete PTHrP, which then acts on the PTH1 receptor to increase bone resorption and renal calcium reabsorption. By inducing the production of high-affinity neutralizing antibodies against PTHrP, the vaccine aims to sequester the protein and prevent its pathological signaling, thereby normalizing serum calcium levels in cancer patients.
