Drug intelligence / Profile preview
Anti-PTX3 monoclonal antibodies are therapeutic candidates designed to target and neutralize pentraxin 3 (PTX3), a soluble pattern recognition receptor and acute-phase protein involved in innate immunity, inflammation, and tissue remodeling. In the tumor microenvironment, PTX3 is primarily secreted by cancer-associated fibroblasts and promotes tumor progression and immunosuppression by driving the polarization of macrophages toward a protumoral, M2-like phenotype. By blocking PTX3, these monoclonal antibodies inhibit the PTX3/CD44 interaction, suppress M2-like macrophage activity, and restore anti-tumor immunity by increasing the infiltration and activity of cytotoxic CD8+ T cells. Preclinical studies, such as those evaluating the neutralizing antibody WHC-001, have demonstrated the potential of anti-PTX3 monoclonal antibodies to reduce stroma-mediated tumor growth in colon cancer and triple-negative breast cancer, as well as to mitigate tissue fibrosis.
