Drug intelligence / Profile preview
Anti-PvAMA1 human monoclonal antibodies are a panel of human monoclonal antibodies (including the lead candidate humAb 826827) that target the Apical Membrane Antigen 1 (PvAMA1) of *Plasmodium vivax*. PvAMA1 is an essential parasite ligand involved in both sporozoite invasion of hepatocytes (pre-erythrocytic stage) and merozoite invasion of reticulocytes (blood stage). By binding to PvAMA1 with high affinity, these antibodies block its interaction with its receptor, rhoptry neck protein 2 (RON2), thereby preventing host cell invasion. Developed by a collaborative research network including Case Western Reserve University, the Walter and Eliza Hall Institute, and the Burnet Institute, these antibodies (particularly humAb 826827) have demonstrated potent strain-transcendent inhibition of *P. vivax* infection in vitro and in vivo, representing a promising therapeutic and prophylactic strategy against vivax malaria.
