Overview

Anti-RAGE antibodies are a class of investigational monoclonal antibodies designed to target the Receptor for Advanced Glycation End Products (RAGE), a multi-ligand pattern-recognition receptor and member of the immunoglobulin superfamily. RAGE is implicated in the pathogenesis of various chronic inflammatory and metabolic diseases, including Alzheimer's disease, diabetes, atherosclerosis, and several cancers, where its overexpression drives pro-inflammatory and pro-survival signaling. By neutralizing RAGE or blocking its interaction with ligands such as advanced glycation end products (AGEs), HMGB1, and S100 proteins, these antibodies aim to mitigate tissue damage and inhibit tumor progression. Research programs include humanized antibodies for inflammatory conditions and antibody-drug conjugates (ADCs) like RBGO1, which are being explored for the treatment of RAGE-overexpressing gynecological malignancies such as endometrial and ovarian cancer.
