Drug intelligence / Profile preview
Anti-RANKL refers to a class of monoclonal antibodies and therapeutic agents designed to target and neutralize Receptor Activator of Nuclear Factor κB Ligand (RANKL). RANKL is a key cytokine in bone metabolism that drives the differentiation, activation, and survival of osteoclasts, which are the cells responsible for bone resorption. By binding to RANKL, anti-RANKL agents prevent it from interacting with its receptor, RANK, on osteoclast precursors and mature osteoclasts. This inhibition halts osteoclastogenesis, reduces bone resorption, and increases bone mineral density. The most prominent clinical representative of this class is denosumab (marketed as Prolia and Xgeva), which is widely approved for the treatment of postmenopausal osteoporosis, treatment-induced bone loss, giant cell tumor of bone, and bone metastases from solid tumors. Preclinical research also frequently utilizes various anti-RANKL antibodies (such as clone IK22/5) to study bone remodeling, fracture repair, and osteo-immune interactions.
