Anti-RelA siRNA is a small interfering RNA (siRNA) designed to specifically silence the expression of the RelA subunit of nuclear factor kappa B (NF-κB), a key transcription factor involved in inflammatory and immune responses. By targeting and degrading RelA mRNA via the RNA-induced silencing complex (RISC), anti-RelA siRNA inhibits production of pro-inflammatory cytokines such as TNF-α and IL-6. This approach has been investigated primarily for topical treatment of atopic dermatitis, where encapsulation in flexible DOPE/CHEMS liposomes enhances skin penetration and therapeutic efficacy. The mechanism relies on sequence-specific mRNA degradation post-transcriptionally, offering high specificity with minimal off-target effects[1][4][6]. Anti-RelA siRNA-based therapies are still experimental and have not reached clinical approval.
