Overview

anti-RGS2 siRNA is a small interfering RNA (siRNA) designed to silence the expression of the Regulator of G protein signaling 2 (RGS2) gene. RGS2 is a critical modulator of G protein-coupled receptor (GPCR) signaling, specifically acting as a GTPase-activating protein (GAP) to terminate Gαq-mediated bronchoconstrictive signals. In research settings, such as studies on human airway smooth muscle (HASM) cells, anti-RGS2 siRNA is used to model the effects of RGS2 deficiency, which is associated with a hypercontractile phenotype and increased asthma exacerbation rates. By reducing RGS2 protein levels, the siRNA enhances intracellular calcium flux and contractile responses to agonists like histamine and methacholine, thereby validating the causal relationship between reduced RGS2 expression and airway hyperresponsiveness.
