**Anti-ROR1 CAR NK Cells** are an investigational cell therapy consisting of natural killer (NK) cells genetically engineered to express a chimeric antigen receptor (CAR) targeting receptor tyrosine kinase-like orphan receptor 1 (ROR1), a tumor-associated antigen overexpressed in various solid tumors and hematological malignancies. The CAR redirects NK cells to recognize and lyse ROR1-positive cancer cells in an MHC-independent manner, leveraging NK cells' innate cytotoxicity, enhanced persistence, and reduced risk of cytokine release syndrome compared to CAR-T cells. Developed for challenging indications like neuroblastoma, glioblastoma, and other ROR1-expressing solid tumors, this therapy is being explored in preclinical studies and early clinical combinations, such as with cytokine-secreting oncolytic viruses or IL-15 superagonists like N-803, to overcome immunosuppressive tumor microenvironments.[2][3][4]
