anti-ROR1 CAR T cells are autologous or allogeneic T cells engineered to express a chimeric antigen receptor (CAR) that specifically recognizes receptor tyrosine kinase-like orphan receptor 1 (ROR1), a transmembrane protein aberrantly expressed on various hematologic and solid tumor cells, but generally absent from normal adult tissues. The engineered CAR T cells, using scFv sequences such as those derived from monoclonal antibodies like R12, R11, or cirmtuzumab, enable these T cells to specifically bind ROR1 and mediate targeted cytotoxicity against ROR1-positive tumor cells. Mechanism of action: The CAR, comprising an extracellular antigen-recognition domain (anti-ROR1 scFv), hinge region, transmembrane domain, one or more intracellular co-stimulatory domains (most commonly CD28 or 4-1BB), and a T-cell signaling domain (CD3ζ), redirects T-cell activity toward ROR1-expressing malignant cells, leading to their direct killing. anti-ROR1 CAR T cells are in early clinical and preclinical development for hematologic malignancies (e.g., chronic lymphocytic leukemia, mantle cell lymphoma) and solid tumors (e.g., breast cancer, lung cancer, other carcinomas).
