An anti-ROR1 monoclonal antibody is a type of targeted biologic therapy designed to bind to Receptor Tyrosine Kinase-Like Orphan Receptor 1 (ROR1), an oncofetal cell-surface antigen. ROR1 is highly expressed during embryonic development and aberrantly overexpressed on various hematological malignancies (such as chronic lymphocytic leukemia and mantle cell lymphoma) and solid tumors (including breast, ovarian, and lung cancers), while being largely absent from normal adult tissues. By binding to ROR1, these monoclonal antibodies can block non-canonical Wnt5a-induced ROR1 signaling, which is implicated in tumor cell survival, proliferation, metastasis, and self-renewal. Additionally, some anti-ROR1 antibodies are engineered to enhance immune-mediated effector functions, such as antibody-dependent cellular cytotoxicity (ADCC). Prominent examples of clinical-stage candidates in this class include zilovertamab (formerly cirmtuzumab) and PBA-0405.
