Overview

Anti-SCUBE3 monoclonal antibody is a therapeutic biologic designed to target and neutralize the secretory protein SCUBE3 (Signal peptide, CUB domain and EGF-like domain-containing protein 3). Research indicates that SCUBE3 acts as an oncoprotein in triple-negative breast cancer (TNBC) by functioning as an extracellular ligand for receptors such as EGFR, CALR, and TGFβRI/II, which in turn activates pro-growth signaling pathways including FOXR2 and c-Myc. Furthermore, SCUBE3 contributes to an immunosuppressive tumor microenvironment by inhibiting antigen presentation through the Myc-DNMT1-IRF1 axis. The antibody, which has been engineered with an LALAPG mutation in the heavy chain to optimize its profile, has demonstrated preclinical efficacy in reducing tumor growth and overcoming therapy resistance in TNBC and ovarian cancer models.
