Drug intelligence / Profile preview
Anti-Sema4A neutralizing antibody refers to monoclonal or polyclonal antibodies designed to bind and neutralize semaphorin-4A (Sema4A), a transmembrane glycoprotein involved in immune regulation, T-cell activation, and angiogenesis. Sema4A is elevated in several autoimmune and inflammatory conditions, including psoriatic arthritis (PsA), systemic sclerosis (SSc), rheumatoid arthritis (RA), and multiple sclerosis (MS). By blocking the interaction of Sema4A with its receptors (such as Plexin-B2, Plexin-D1, and Neuropilin-1), these neutralizing antibodies inhibit Th17 and Th2 cell differentiation, reduce the production of inflammatory cytokines (such as IL-17), and attenuate tissue fibrosis and inflammation. Preclinical studies have demonstrated that anti-Sema4A neutralizing antibodies can reduce the severity of collagen-induced arthritis (CIA) and bleomycin-induced systemic sclerosis in animal models.
