Overview

The anti-SEMA7A monoclonal antibody (specifically clone SmAbH1) is a preclinical-stage biologic therapeutic that targets Semaphorin 7A (SEMA7A), a GPI-anchored glycoprotein. Developed by researchers at the University of Colorado Anschutz Medical Campus, SmAbH1 is being investigated as a novel treatment for breast cancer, particularly estrogen receptor-positive (ER+) and triple-negative breast cancers (TNBC) where SEMA7A is overexpressed. SEMA7A overexpression is associated with poor prognosis, metastasis, and resistance to endocrine therapy. SmAbH1 works by blocking SEMA7A signaling through its receptors, such as integrins (including integrin beta-1 and beta-4), thereby inhibiting downstream pro-survival AKT signaling, reducing tumor cell viability, and reversing therapeutic resistance. It has demonstrated preclinical efficacy both as a monotherapy and in combination with endocrine therapies (like fulvestrant or tamoxifen) and PI3K inhibitors.
