The anti-sialyl-Tn antibody-drug conjugate (ADC) is a preclinical biologic therapeutic developed by Siamab Therapeutics. It consists of a humanized monoclonal antibody targeting the tumor-associated carbohydrate antigen (TACA) Sialyl-Tn (STn), also known as CD175s or the Sialyl-Thomsen-nouveau antigen. The antibody is conjugated to the cytotoxic payload monomethyl auristatin E (MMAE), a potent anti-mitotic agent that inhibits tubulin polymerization. STn is highly expressed on the surface of various epithelial cancers, including ovarian, colorectal, breast, prostate, and pancreatic tumors, but is rarely found in normal adult tissues. The ADC is designed to selectively deliver MMAE to STn-expressing cancer cells, exploiting the tumor-specificity of the antigen to minimize off-target toxicity and potentially overcome immune suppression and chemoresistance associated with STn expression.
