Anti-Siglec-15 antibody-GM-CSF chimera is a recombinant fusion protein designed for cancer immunotherapy. It combines an antibody targeting Siglec-15—a sialic acid-binding immunoglobulin-like lectin implicated in tumor-associated macrophage (TAM)-mediated immunosuppression—with granulocyte-macrophage colony-stimulating factor (GM-CSF), a cytokine that promotes immune cell activation. The drug acts by blocking the immune checkpoint molecule Siglec-15 on TAMs and delivering GM-CSF specifically to the tumor microenvironment, thereby reprogramming TAMs from an immunosuppressive to a pro-inflammatory phenotype. This enhances antigen presentation and T cell activation within tumors. In preclinical models, this approach demonstrated superior suppression of solid tumor progression compared to either component alone, with improved localization to Siglec-15-positive cells and reduced off-target effects[1][2][3][4].
