Anti-Siglec-6 antibodies are a class of investigational biologics that target Sialic acid-binding Ig-like lectin 6 (Siglec-6, also known as CD327), an inhibitory receptor primarily expressed on mast cells, B cells, and plasmacytoid dendritic cells (pDCs). In malignancies such as chronic lymphocytic leukemia (CLL) and multiple myeloma (MM), Siglec-6 is upregulated and acts as an immune checkpoint, facilitating tumor evasion by transmitting inhibitory signals upon binding to sialylated glycans. Therapeutic antibodies against Siglec-6 aim to block these inhibitory interactions, thereby restoring immune cell activation and promoting the killing of malignant cells. Research is also exploring anti-Siglec-6 antibodies for the treatment of mast cell-mediated allergic and inflammatory disorders, leveraging the receptor's restricted expression pattern to achieve high therapeutic selectivity.
