Anti-Siglec-6 CAR-T cell therapy is an autologous chimeric antigen receptor (CAR) T-cell immunotherapy developed by Xuzhou Medical University for the treatment of relapsed and refractory acute myeloid leukemia (AML). The therapy involves genetically engineering a patient's T cells to express a CAR that targets Siglec-6 (sialic acid-binding immunoglobulin-like lectin 6), using a targeting domain derived from the human monoclonal antibody JML-1. Siglec-6 is highly expressed on AML blasts and leukemic stem cells but is absent on normal hematopoietic stem and progenitor cells (HSPCs), potentially reducing off-target myelotoxicity. The therapy is currently undergoing Phase 1/2 clinical evaluation to assess its safety and efficacy in patients with advanced AML.
