Anti-Siglec-E antibody is a monoclonal antibody that targets Sialic acid-binding Ig-like lectin E (Siglec-E), an inhibitory receptor found on murine myeloid cells, including microglia and macrophages. Siglec-E is the mouse functional homolog of human Siglec-9 and serves as an innate immune checkpoint. It recognizes sialic acid ligands on the surface of cells; in the tumor microenvironment, cancer cells exploit this interaction to evade immune clearance by sending 'don't eat me' signals. By blocking the Siglec-E receptor, the antibody prevents these inhibitory signals, thereby promoting the phagocytosis of tumor cells by microglia. Research, such as that presented by the University Hospital Basel, utilizes anti-Siglec-E antibodies in preclinical models of glioblastoma to investigate the therapeutic potential of targeting the sialic acid-Siglec axis to enhance anti-tumor innate immunity.
