Overview

Anti-SIRPα antibodies are a class of monoclonal antibodies designed to target Signal Regulatory Protein Alpha (SIRPα), an inhibitory receptor primarily expressed on myeloid cells such as macrophages, monocytes, and dendritic cells. SIRPα serves as a critical immune checkpoint by interacting with CD47 (the "don't eat me" signal) on the surface of other cells, which inhibits phagocytosis. By binding to SIRPα and blocking this interaction, these antibodies relieve the inhibition of myeloid cells, thereby promoting the phagocytic clearance of target cells, including cancer cells and pathogenic stromal cells. In oncology, anti-SIRPα antibodies are being investigated to enhance antibody-dependent cellular phagocytosis (ADCP) and stimulate adaptive immunity through improved antigen cross-presentation. Beyond cancer, they are also being explored for their potential to treat fibrostenotic complications in inflammatory bowel diseases, such as Crohn's disease, by modulating the crosstalk between macrophages and pro-fibrotic stromal cells.
