The anti-SLC7A11 antibody is a humanized monoclonal antibody designed to target the cystine/glutamate antiporter SLC7A11 (also known as xCT). SLC7A11 has been identified as an immunometabolic checkpoint that is significantly upregulated in exhausted CD8+ T cells within the tumor microenvironment. By blocking cystine uptake, the antibody triggers an integrated stress response characterized by the activation of the GCN2-eIF2α-ATF4 pathway, which in turn transcriptionally upregulates the glutamine transporter SLC1A5. This metabolic reprogramming increases glutamine uptake and mTORC1 signaling, shifting T cell metabolism toward mitochondrial oxidative phosphorylation (OXPHOS) and increasing ATP production. Preclinical studies indicate that treatment with this antibody alleviates CD8+ T cell exhaustion, promotes the expansion of stem-like memory T cells (TSCM), and enhances the anti-tumor potency of CAR-T cell therapies and immune checkpoint blockades in solid tumors, including colorectal and breast cancer models.
