The anti-SLC7A11 monoclonal antibody is a humanized monoclonal antibody targeting solute carrier family 7 member 11 (SLC7A11, also known as xCT), a cystine/glutamate antiporter overexpressed in various solid tumors. Developed by researchers at Peking University First Hospital, the antibody blocks SLC7A11-mediated cystine uptake, which alleviates CD8+ T cell exhaustion and promotes the expansion of stem-like memory T (Tscm) cells. Mechanistically, SLC7A11 inhibition activates the GCN2-eIF2α-SLC1A5 signaling axis, increasing glutamine uptake and enhancing mitochondrial oxidative phosphorylation and bioenergetic fitness. In preclinical models, treatment with the anti-SLC7A11 antibody significantly enhances the antitumor efficacy of CAR-T cell therapies and synergizes with immune checkpoint blockade.
