Overview

Anti-SPP1 antibodies are a class of monoclonal antibodies designed to neutralize Secreted Phosphoprotein 1 (SPP1), also known as Osteopontin (OPN). SPP1 is a multifunctional matricellular protein that plays a pivotal role in immune regulation, inflammation, and tissue remodeling. In the context of oncology, SPP1 is highly expressed by a specific subset of tumor-associated macrophages (TAMs), where it acts as a potent immunosuppressive factor. By binding to receptors such as CD44 and various integrins (e.g., αvβ3), SPP1 promotes T-cell exclusion, inhibits the activation of cytotoxic T lymphocytes, and facilitates tumor metastasis and epithelial-to-mesenchymal transition (EMT). Therapeutic anti-SPP1 antibodies aim to disrupt these signaling pathways to reverse immune evasion and enhance the efficacy of checkpoint inhibitors. Additionally, these antibodies are being explored for the treatment of fibrotic diseases, where SPP1 contributes to myofibroblast activation and extracellular matrix deposition.
