Anti-TA-MUC1 antibody refers to a monoclonal antibody that specifically targets tumor-associated mucin-1 (TA-MUC1), a transmembrane glycoprotein that is aberrantly glycosylated and overexpressed in various epithelial cancers, including lung, breast, and bladder cancers. Unlike normal MUC1, TA-MUC1 features truncated O-glycans (such as Tn and sialyl-Tn antigens) that expose unique glycopeptide epitopes on the protein backbone. These antibodies are primarily utilized as the targeting component in therapeutic modalities such as antibody-drug conjugates (ADCs), including Daiichi Sankyo's DS-3939a. By recognizing both the glycan and backbone peptide moiety of TA-MUC1, these antibodies facilitate the selective delivery of cytotoxic payloads (like the DNA topoisomerase I inhibitor DXd) to malignant cells while sparing normal tissues. Preclinical studies indicate that while the parental antibody may lack potent standalone anti-tumor activity, it provides the high specificity required for targeted cancer therapy.
