Overview

Anti-TACI CAR T cells are an experimental adoptive cell therapy designed to treat multiple myeloma by targeting the transmembrane activator and CAML interactor (TACI; TNFRSF13B). Developed by researchers at Massachusetts General Hospital, these second-generation chimeric antigen receptor (CAR) T cells utilize a novel single-chain variable fragment (scFv) derived from immunized mice. TACI is a cell surface receptor expressed on malignant plasma cells, and its targeting is intended to provide a therapeutic alternative or complement to BCMA-targeted therapies, particularly to address BCMA-negative relapses. Preclinical data indicate that anti-TACI CAR T cells demonstrate potent in vitro cytotoxicity and in vivo anti-tumor activity in xenograft models of multiple myeloma.
