Anti-TACI dual CAR T cells are an experimental cellular immunotherapy consisting of chimeric antigen receptor (CAR) T cells engineered with a dual-specific tandem scFv construct targeting both Transmembrane Activator and CAML Interactor (TACI) and B Cell Maturation Antigen (BCMA). This biologic is designed to treat multiple myeloma, specifically addressing the challenge of antigen escape or loss that often occurs after treatment with BCMA-directed therapies. By co-targeting TACI, these cells aim to maintain efficacy against myeloma cells that have downregulated BCMA. Additionally, the construct is designed to potentially offer a safer neurotoxicity profile compared to high-affinity BCMA-only CARs, as TACI RNA expression is limited in the basal ganglia. The research describing this construct was published in Nature Communications in November 2023.
