anti-TAG72-IL-2 is a fully humanized immunocytokine that combines an antibody targeting tumor-associated glycoprotein 72 (TAG-72) with interleukin-2 (IL-2). TAG-72 is a cell surface antigen highly expressed in various epithelial-derived cancers, including colorectal cancer. The drug is designed to deliver IL-2 directly to TAG-72–expressing tumor cells, thereby stimulating local immune responses and enhancing anti-tumor activity. Mechanistically, the antibody component binds specifically to TAG-72 on tumor cells, while the IL-2 moiety activates immune effector cells such as T cells and natural killer (NK) cells within the tumor microenvironment. Preclinical studies have demonstrated significant tumor reduction and even cures in mouse models of TAG‑72–positive colorectal tumors[1]. The approach aims to maximize immune-mediated destruction of cancer while minimizing systemic toxicity associated with high-dose IL‑2 therapy.
