Overview

Anti-TCRVβ2 CAR-iNKT cells are an experimental allogeneic cell therapy consisting of invariant natural killer T (iNKT) cells engineered to express a chimeric antigen receptor (CAR) targeting the TCRVβ2 subunit of the T-cell receptor. Developed by the Karadimitris Lab at Imperial College London, this therapy is designed to treat T-cell malignancies, particularly Adult T-cell leukaemia/lymphoma (ATL) and other peripheral T-cell lymphomas (TCL) that clonally express the TCRVβ2 chain. By targeting a specific TCRVβ family, the therapy aims to eliminate malignant clones while sparing the majority of the healthy T-cell repertoire. The use of iNKT cells as effectors facilitates an "off-the-shelf" allogeneic approach because these cells do not induce graft-versus-host disease (GVHD).
