Drug intelligence / Profile preview
Anti-TFPI antibodies are a class of monoclonal antibodies designed to target and inhibit tissue factor pathway inhibitor (TFPI). TFPI is an endogenous anticoagulant that regulates the initiation of blood coagulation by blocking the tissue factor-factor VIIa (TF-FVIIa) complex and factor Xa. By neutralizing TFPI, these antibodies enhance thrombin generation through the extrinsic pathway, effectively bypassing the need for coagulation factors VIII or IX. This mechanism makes them valuable as non-factor prophylactic therapies to prevent bleeding episodes in patients with hemophilia A or B, both with and without inhibitors. Key clinical-stage and approved candidates in this class include Pfizer's marstacimab (Hympavzi) and Novo Nordisk's concizumab (Alhemo), which are administered subcutaneously.
