The anti-TfR nanoconjugate is a target-specific generic platform descriptor for nanoconjugate drug delivery systems that incorporate an anti-transferrin receptor (TfR) monoclonal antibody as a targeting moiety. Typically built on a biodegradable poly(β-L-malic acid) (PMLA) nanoplatform (often referred to as Polycefin), these constructs are designed to cross the blood-brain barrier (BBB) or blood-tumor barrier (BTB) via receptor-mediated transcytosis. The anti-TfR antibody component targets the transferrin receptor, which is overexpressed on tumor vasculature and tumor cells. Depending on the specific variant, the nanoconjugate can be loaded with various therapeutic payloads, such as antisense oligonucleotides (e.g., targeting EGFR or laminin-411), other tumor-targeting antibodies (e.g., mAb 2C5), or chemotherapeutic agents (e.g., temozolomide). These platforms have been investigated in preclinical research by Cedars-Sinai Medical Center and its spinout Arrogene for the treatment of brain tumors (such as glioma and glioblastoma) and triple-negative breast cancer.
