Overview

The anti-TfR1 multivalent peptide is a preclinical therapeutic agent developed by Genentech that targets the Transferrin Receptor (TfR1). Unlike traditional antibody-based degraders (PROTABs) that recruit E3 ligases, this multimeric peptide modality induces the clearance and degradation of TfR1 from the cell surface through target clustering. TfR1 is frequently upregulated in various cancers, including colorectal cancer (CRC), to support increased iron uptake. By clustering TfR1, the peptide induces target degradation and subsequent cell viability defects. This approach represents a novel targeted protein degradation (TPD) strategy that leverages target clustering rather than specific ligase recruitment to affect cell fitness across multiple cancer indications.
